I have Schwalbe Big Apples 700 x 50 on one of my bikes. I suppose I am not going to win a race with the bike, but then, I don't race.
Since setting up the bike, I have used it for two weekend camping trips - both times road about 200 miles over three days. The tires are comfortable and ride very well. Rolling resistance is very low. The bike is heavy, but that has more to do with the extra wide Berthoud stainless steel fenders, Rohloff hub, and Cetma 5 rail up front. In fact, I like it so much, I am toying with selling my current long haul camper - a fully restored Trek 728 which takes max 35s and is probably better off with 32s.
