Hi Careina Initially I was sure they were protein crystals, principally because you only have NaCl, Hepes and TCEP in your buffer. And then you said they are hard. If you try to press them against the bottom of the well, they should easily break if they are made of proteins. Once they did not diffract, I think it is something you should try with the remaining ones. Alternatively, if you have a source of UV light coupled to a camera, they should “light up”.
