Sounds like the configuration may be hosed. Have you tried reconfiguring LyX? If that doesn't work, you might try deleting the local LyX subdirectory (probably C:\Documents and Settings\\Application Data\lyx16, but your mileage may vary), backing up any modified layouts etc. you might have there,
