At 300 lbs I'd recommend 48 spokes rear and 36 up front.  650B or 559  
rather than 622 seems like a good choice.  Ideally I'd also recommend  
a wide spaced tandem rear hub (145 mm instead of 130 or 135) to  
minimize dish and maximize the stability of the rear wheel.
