Yes, you can. I do it all the time with Vista and
previously did it all the time with XP.

Verified. Just tried
* renaming my LilyPond directory (on Win XP) containing 2.13.26
-> lilypond -version failed (command not found)
* installing LilyPond 2.13.28
-> new LilyPond directory created by installer
* renaming this new directory
- > lilypond -version failed
* When renaming one of the two to LilyPond
-> lilypond -version shows the respective version number
Well, this was quite simple for launching such a long thread ...
