It makes a big difference on how the particular piece is loaded.  If it is
loaded only in compression like the vertical spacers in the spars you could
actually make the piece stronger.  If you do it in a piece that is loaded in
tension or bending it is a totally different story.

You didn't say how big the hole was going to be or what part of the
structure it is located in.  All relevent for meaningful answer.  The
front vertical stab spar has a hole drilled through the middle for
the elevator cable and nothing glued in the hole.

I'd need more info to even give you an uneducated answer.
