We generally only use Mitegen In-situ-1 plates with optimisation conditions
dispensed with the Formulator not initial screens. However, our Art-Robbins
Pheonix (once I repair it) can dispense into these trays from a deep well block
(as would the Hydra it replaced as they share a similart head). It would
probably work better with the finer flexible needles given the small target. We
don't dispense the protein, just aliquot the screen into the reservoir and then
move to the NT8 or Mosquito.

We have also tried dispensing deep well blocks from tube format using larger
more generic liquid handling robots in our high throughput facility. This works
fairly well but not all screens behave due to a mix of variable viscosity and
surface tension across the conditions.

Hope this is of interest.

Andy

External Sender: Use caution.

We use the Formulator from Formulatrix for mixing cocktails. We use it to mix
96 well deep well blocks for preparing the 1536 high-throughput screen we use
in the National Crystallization Center at HWI, as well as for optimization
plates. I'm not sure if any of the others can dispense into deep well blocks –
if anyone uses another robot to do so, please let me know!

I echo Halina's request – does anyone make use of a robot to set up Mitegen in
situ plates?

Warmly,

Sarah

Dear crystallographers,
we are thinking about buying a new robot for design and pipetting screens. What
instruments do you use ? Any recommendations or experiences you would like to
share ?

Kind regards,
Ulrike
