I daily use Tev protease for a ration 1:20 (Tev-protein) but the optimal activity of TEV is at 31°C for 1 hour, if there is not the case, you can put the clivage reaction at 4°C for ON. In some cases, cleavage is not at 100% (SDS-PAGE control) and you need to put reaction for more time (2 days approximately). In lectures, it is described that they use a differents ratio (1:5, 1:10, 1:20) but i think the efficient way is to test differents ratio with a small volume reaction (20uL for example).
