What is your ratio of protein to ligand?  You might want to try 1:3, 1:5 
or 1:10 ratios of protein : ligand.  This can make a HUGE difference. 
There are instances where the organic solvents (DMSO, EtOH) used to 
dissolve the ligands interfere with the crystallization process.  In this 
instance I  tried the following:

put dry cmpd in the crystallization drop with the protein  (does not 
matter if it does not totally dissolve)

added the dry cmpd to the protein solution & put on a LabQuake shaker 
overnight with gentle rotation.  Centrifuge the next morning & set up your 
crystallization screens.

[ccp4bb] Substrate(water insoluble) Soaking






 
 
 
 Hi All,
     I am now soaking two substrates to the crystal.But one is soluble in 
ethanol and one is soluble in DMSO. After co-crystallization and soaking, 
no substrate found in the structure.
     Does anybody has the experience in dealing this problem?
     Any comments or suggestions would be highly welcome.
