Here are a few personal opinions,  (although my assumptions may be
wrong or out of date):

GAE
1. web2py targets the GAE platform and a lot of effort is put into
this brand of 'No-Sql'.  The web2py lead developer has access to
forthcoming enhancements and so it should function very well in the
future too.
2. GAE does not have SQL databases yet, but that will change soon. We
do not know exactly what to expect yet of the Sql stuff,  but I
imagine it will be good. There may be some delay for web2py to offer
an adapter for the new DB, but I imagine that would be quick.

EC2
1. EC2 can be used for scalable SQL server hosting services and has
super-fast access to S3 storage at low storage rates.
2. EC2 is ideal for scaling applications of very large enterprises. It
is a little more expensive than other 'cloud' providers and not as
easy to use.
3. web2py does not target the No-Sql SimpleDB database.

Question is: do you want No-Sql or Sql?  If the former,  then choose
GAE.  If the latter and you can wait,  then see what the Google Sql
offers.  If you can't wait,  then use EC2.  However, rather than use
EC2,  I would recommend considering alternatives due to ease of use,
and, to a lesser degree, cost.  Check out Webfaction for quick
installation of web2py.  I personally use vps.net and am very happy :)
