i use postscreen, clamav-milter ( sanesecurity) , opendkim opendmarc policyd-spf spamass-milter, postgrey ( only selective ) and as well lots of other restictions so there are a few "best practises" how to setup but at the very end you have to analyse your logs what might be best at your site, cause everyone has its own "unwanted stuff". Greylisting is not a big factor in my setups since years, too many bots have full smtp engines implemented which pass greylisting. Contentfilter like spamassassin have high "costs", so make sure to reject as most as possible by other stuff before pass to spamassassin.
