Bonjour,
I want to make a poster using xelatex and bearmerposter package. For the
main text, it is required to use Arial fonts at 44pt with a baselineskip
at 60pt.
Is there a way to fix these sizes in a \newfontfamily command?

No. Just use the NFSS2 means (\fontsize{44}{60pt}\selectfont or such)!
