Probably by removing the blank line at the top of your spool file that
somehow got there and which makes the mailbox no longer an mbox. We get
this sort of question a lot; I bet it would be easy to find

I can duplicate your problem if I put some garbage like as the first
line of my mail box file. That is, file reports a mail box but mutt
complains it is not.

Don't do that. Rather, you should check whether
/usr/local/bin/mutt_dotlock is setgid mail.
