Dear colleagues,

We have a PhD student position available, for structural and functional studies 
of bacterial Type 4 Secretion Systems.
We are looking for an enthusiastic and highly motivated individual who has an 
interest in large protein machineries. We use biochemistry, molecular biology, 
x-ray crystallography and cryo-EM to study Type 4 Secretion Systems, mainly of 
Gram-positive origin.

The Berntsson lab is part of the integrated structural biology network at Umeå 
University, and from the 1st of December also part of the Wallenberg Center for 
Molecular Medicine. We are well equipped with state of the art technologies, 
both for crystallography and EM, including a nanodrop crystallisation robots, 
Formulatrix crystallisation hotel, an EM facility with Titan Krios and Scios 
Dualbeam for FIB-milling, as well as other strong facilities.

[ccp4bb] cryo suggestions for lithium sulphate xt condition containing 1.8 - 2 M Li2SO4

Dear all,

I have looked into the archive and I've tried to collect suggestions for
cryo protecting crystals that grew in 1.8 M Lithium sulphate containing
mother liquor or 2M.

What would make more sense? Transferring it into a cryo solution that
consists of mother liquor and a higher concentration of Lithium sulphate?

Or, would this concentration of Lithium sulphate be enough as a
cryo-protectant and after slight dehydration I could flash freeze them?

I will start my group at the Max F. Perutz Laboratories (MFPL) in Vienna in 
March 2019. I am currently recruiting a PhD student through the Vienna 
BioCenter PhD Programm (see below).
Note: Only application through the Vienna BioCenter PhD program can be

Project: My group is interested in unraveling the molecular mechanisms that 
allow the establishment of a robust RNA interference (RNAi) response in which 
small RNAs are used to silence the expression of genes. We are particularly 
interested in understanding how the nuclear RNAi pathway leads to 
heterochromatin formation. We will address these questions by reconstituting 
and characterizing the protein complexes involved in these pathways both 
biochemically and structurally. For the structural characterization, we employ 
an integrative approach by combining X-ray crystallography and single-particle 
electron microscopy. This will be complemented by biochemical approaches 
(enzyme activity assays) and biophysical assays (isothermal titration 
calorimetry, analytical ultracentrifugation, and spectroscopic methods). For

Hi Almu,

In my hands, 2M LiSO4 is not enough.

If your crystals cannot survive transfer to higher concentrations, transfer 
them to 1.8 M NaMalonate at the same pH and then transfer or add aliquots of Na 
Malonate stock at higher concentration until the drop has about 3.5 - 3.8 M Na 
Malonate. The optimal final concentration may vary from system to system. This 
protocol has worked for us many times.

We are opening one postdoctoral position at the Brazilian Biosciences National 
Laboratory - LNBio (http://lnbio.cnpem.br/) in Campinas – Sao Paulo in Dr. 
Andrea Dessen’s group. The position involves: (1) fractionation and 
purification of bacterial secondary metabolites, as well as (2) testing for 
molecule recognition of bacterial cell wall synthesis proteins via 
crystallography. The successful candidate could start in the post during the 
first semester of 2019. The contract is for two years, and there is the 
possibility of an extension.
Candidates should have a recent PhD degree Biochemistry, Structural Biology, 
Chemistry or related disciplines and have published at least one research 
article in an international peer-reviewed journal as first author. The 
successful candidate should have practical knowledge in biochemistry and 
protein structure determination. Demonstrated expertise in one of the following 
areas would be advantageous: natural product purification and chromatographical

[ccp4bb] Winter School of Cryo-EM Workshop at Arizona State University (ASU)

Dear everyone,

We are pleased to announce the Cryo-EM Workshop of the High-Resolution TEM
Winter School, which will be held on December 11-14, 2018 at Arizona State
University (ASU) Tempe campus.  The HRTEM Winter School at ASU has a long
history in educating and training TEM scientists, and we have been
expanding our interests and courses to the biological applications,
especially the cryo-EM methods.

The goal of the Cryo-EM Workshop is to introduce the concepts and
applications of the biological cryo-EM.  We will have four guest speakers
coming to join our workshop: Dr. James Evans (Pacific Northwest National
Laboratory), Dr. Tim Grant (HHMI Janelia Farms), Dr. Axel Brilot (UCSF),
and Dr. Thomas Tomasaik (University of Arizona).  Participants will learn
the complete workflow and advanced techniques used for biological cryo-EM
through lectures, demos, and hands-on experiments.  No previous experience
is required.  Also, it will be a great opportunity for the participants to
learn the cryo-EM principles while working along with their samples.
Therefore, we are welcoming to receive the proposals from who are
interested in bringing their samples and processing their data on site.
