[ccp4bb] on modification the Cbeta of Ser
Dear All, Can anyone tell me how to change the position of Cbeta of Ser by Coot? Cheers, Dialing
Re: [ccp4bb] [OFF-TOPIC] Site-Directed Mutagenesis
Hi Fred, While you are working on the optimization of your PCR, I strongly suggest you to read our paper on partial overlapping primer design from a link below, which will surely ease your mutagenesis: http://www.ncbi.nlm.nih.gov/pubmed/21530481 I would be happy to answer your questions should you have. From quick reading most of the answers, the following seems to be a consensus: 1) Do not concentrate your PCR product; 2) Too much DNA and/or impurities like salts or whatever can inhibits transformation; 3) Purify your PCR product before transformation if possible or use 3 of 4 microL of it. This is more or less the amount of DNA showed in the uploaded image. Kind regards, Fred P.S.: I'll let you know the results. [ccp4bb] off-topic:schematic representations for secondary structure
Dear memebers, Thank you all firsit for the helps on my previous post about sequence alignment. They are really useful! Apologize for the off-topic question again. I usually use DSSP method to calculate the secondary structures on my sovled structures. I saw a nice schematic representation for secondary structure from PDB database website (please see the attached figure). Does anyone know whether there is a program to draw these schematics? Thank you. King regards, Wenhe <><>
Re: [ccp4bb] off-topic:schematic representations for secondary structure
I believe Procheck generates drawings such as those. It generates PostScript files, and if you need to have (eg) jpg files, a PostScript interpreter, screen capture and there you are (Gimp to select only the areas you're interested in) HTH, Fred. Re: [ccp4bb] Bio Rad "Econo" system
I've been using the Econo system for about 20 years for prep scale purifications. I regularly do gradients using an external gradient maker (something like with Hoefer sells - http://www.hoeferinc.com/product.asp?ProdID=171) that feeds into the pump. It's worked reliably for me and allows the use of cheap columns to boot. Good luck, Arthur On Feb 3, 2012, at 4:08 PM, narayan viswam wrote: > Dear CCP4BB, > > Sorry for this off-topic question.
