I now have exclusively cut-out leather saddles, 3 Selle Anatomica and 1 Brooks Flyer. The cut-out was a huge improvement for me with long ride comfort, and has made my choice of bottom layer mostly irrelevant. However I still err towards shorts/pants that have a gusseted crotch so as to not have a seam between my body and saddle. I will wear a chamois on humid summer recreational rides just to help mitigate sweaty undies/shorts which then translates to damp saddle
