I have used the 2i4 a bit and briefly the Scarlet solo. The 6i6 as I understand it is the 2i4 with 2 more line inputs and digital I/o. Unlike the 2i4 however, the 6i6 requires drivers. (The 2i4 doesn’t and as a result is quite accessible since all I/O task are handled from with in the DAW), and it has physical buttons for switching the line inputs between line and instrument right on the device. It appears the 6i6 doesn’t and you will have to switch between line and instrument in the Focusrite Control software., That being said, Focusrite support is good about Remoting into your system to help you set stuff up if needed, but if you want to record say a guitar or bass through the DI often it can be a pain having to switch via software. That being said, the iOS app worked decently well for stuff like that with the Clarett range and the 6i6 looks to use the same iOS app for control, however you will need sighted assistance to pair the iOS app to the app running on your Mac at least once. This may be blasphemous to some but I have to mention if you don’t need the digital I/O the Behringer U-Phoria 404HD is about $100 cheaper and comparable in sound to the scarlet unit. It has 4 Mic Press which can be also used as line or instrument inputs. The line/instrument level switch, the pad, and phantom power are all physical buttons on the unit and no software or drivers are required, it is very stable and runs rock solid even down at 32 samples when tracking with low latency. The only other caveat is that the 404HD only has one headphone jack where the 6i6 has 2. I have sound examples comparing the two units side by side if interested.
