Days after struggling with this (and just after I sent this email) I discovered the domain cannot be listed in mydestination. I.e., I had to setup it as a virtual domain so virtual_transport takes effect.

My current setting is:

mydestination =
virtual_mailbox_domains = /etc/postfix/vhosts
virtual_transport = lmtp:unix:/tmp/lmtp

(Yes, for testing purposes, I created the socket at /tmp/lmtp).

And vhosts has a list of virtual domains.

Letting mydestination empty is the recommended way to do so? Note that I don't more than one domains, only one, but I do have virtual users.
