That could be the problem. Postfix does not have a single "domains_we_host" file. Instead it has a concept of "address class" of which there are currently four (two of which are virtual), each class having its own "domains we host" file. Things have to be in the right place otherwise there will be 'user unknown' or 'relay access denied' problems.
