so for some reason, postfix can't open a map configuration file.

here

$ ls -l /usr/local/etc/postfix/maps/sql/mailbox
-rw-------  1 root  wheel  400 Jul 14 22:08 mailbox

did you notice that this is only readable by root?

and it works without problems.

And I have -rw-r--r-- 1 root root 165 Sep 29 13:45 virtual-mailbox-maps.cf

what operating system are you running? any selinux/apparmor/... around?
Running SUSE Linux Enterprise 10 SP2 (fully patched) and there's no apparmor or SELinux in the picture.

so what is causing a "permission denied"? doesn't Suse have Apparmor?

don't. this has nothing to do with /vmail.

That's where I've been getting confused, I was told vmail had to have the correct owner and permissions.

don't confuse:
- postfix config files and maps
and
- mailboxes
