IPv6 space is considered low quality from an email perspective, so the answer is yes, receivers are likely to care about missing rDNS. Receivers will care as much if not more that IPv4 if anything is out of place with mail from an IPv6. If you're sending from a IPv6 address, make sure all of your ducks are in a line. That means an SPF that don't list too many ranges, DKIM signing using the 5322.From domain and FCrDNS on your SMTP server.
