A few ways. If you go to the dock (VO-d), then hit right arrow until you get to mail, if you select it it will open it up. I usually just use spotlight, and type in mail, then press enter, or I have some keyboard shortcuts set up for common applications like Mail, Safari, Messages, etc.
There are rules available in preferences for performing certain actions when messages are received. You'd probably want to look there.
