In my experience supporting and developing for many companies seriously working with linux - I'd say redhat(Advanced Server family) and suse/Novell (Suse Linux Enterprise Server family - SLES). If it would be one distro I'd actually point to SuSE because the realy big applications for instance Oracle/DB2 db's and apps that run upon them are certified for SuSE much sooner then they are certified for redhat. It was pretty suprising to me as well but it appears that the relationships between the software giants IBM/Oracle/SAP etc... seems to be better with SuSE/Novell. Another factor is what area in the world or what companies do you work for/with If it's americans you get a more redhat centric approach and in europe you may be stoned alive in the village square for muttering redhat in public :)... I still believe you should try to master both of them although yast (SuSE's configuration tool is excellent and I tend to use it alot and thus eliminate my need to know everything in the file level.(A horrifying proccess I agree :) but - hey I'm realy busy and don't have the time to RTFM like I used to).
