I had some success with using a USB microscope and the python opencv libraries. I was looking for the center of a circle of specific diameter and calculated the distance from this found center to the desired position, then moved by that distance to home the axis. It was a fairly quick-and-dirty test I did, but I was quite encouraged by the results.
