The documentation you cited says

perl -w localize.pl -e -l km-KH=en-US -f khmer.sdf

Note the km-KH=en-US instead of =en-US you gave.

Only entire files from a module are extracted, from localize.sdf and
with a fallback to en-US of the corresponding .src files.

Like written in the documentation you cited:

Now translate the strings in the new created file khmer.sdf.
Merge the strings back:

perl -w localize.pl -m -l km-KH -f khmer.sdf

The translation isn't merge to the .src files, it is merged to the
localize.sdf files, which are taken during the build.
