Hi 白雪慧 Initially I would try to diffract these crystals befofe any optimisation. A microfocus beam should give you some clues. You might find out that you have a super ordered crystal at 1.2 A resolution already, or you crystallized salt or a contaminant.
Further, have you tried seeding? These crystals look ideal for it. I would also reccommend leaving your plates at cold temperature (10 or 4ºC). This will slow down your nucleation events increasing the likelihood of bigger crystals. Best wishes ____________________________________________________ Rafael Marques da Silva PhD Student – Structural Biology University of Leicester Mestre em Física Biomolecular Universidade de São Paulo Bacharel em Ciências Biológicas Universidade Federal de São Carlos phone: +44 07861 273773 "A sorte acompanha uma mente bem treinada" ________________________________________________ ________________________________ De: CCP4 bulletin board <CCP4BB@JISCMAIL.AC.UK> em nome de 白雪慧 <zb20193020...@cau.edu.cn> Enviado: segunda-feira, 16 de dezembro de 2024 08:59 Para: CCP4BB@JISCMAIL.AC.UK <CCP4BB@JISCMAIL.AC.UK> Assunto: [ccp4bb] Crystal optimization Hello, everyone. Thank you for your help earlier.I also found that there is a strange phenomenon, want to see how you optimize. The crystal is a cluster with many nuclei, but regular crystals, and occasionally single crystals, grow out of the holes around the circle. It's in the picture. Have you ever had that happen to you?How to optimize? ________________________________ To unsubscribe from the CCP4BB list, click the following link: https://www.jiscmail.ac.uk/cgi-bin/WA-JISC.exe?SUBED1=CCP4BB&A=1 ######################################################################## To unsubscribe from the CCP4BB list, click the following link: https://www.jiscmail.ac.uk/cgi-bin/WA-JISC.exe?SUBED1=CCP4BB&A=1 This message was issued to members of www.jiscmail.ac.uk/CCP4BB, a mailing list hosted by www.jiscmail.ac.uk, terms & conditions are available at https://www.jiscmail.ac.uk/policyandsecurity/
