Dear All,

             We have recently performed a thermal melt curve analysis using
DSF for a target protein using SYPRO-Orange Dye.
We are extending our studies on the same protein with time-lapse experiment
(ie., RFU Vs Time) at a constant temperature (say 37 degC) using the same
extrinsic SYPRO-Orange Dye.  While RFU's are positive for melt curves,   We
see RFU's in -ve range for the time-lapse curve. We are using the
instrument *RT-PCR (Biorad CFX96).*

            We would be grateful for any suggestions on related literature
or protocol for the time-lapse experimental strategy.
