You can stream the file from the Binary Table to the absolute path of your choice (that you have permissions to), and then read it at your leasure, but if the file is available at build time, it will be a lot cheaper and more reliable to read it then.
Depending on the type of file, you may even be able to use WiX binding variables to get what you want at link time. Never do something at run time you can do at compile or link time.
