you would have to add a new constraint at the database level like this:

        query="ALTER TABLE %s ADD CONSTRAINT %s UNIQUE (amcno,unitid);" 
%(tablename,constraintname)
        result=db.executesql(query)


here a postgresql example to set the new constraint in a controller:

def setconstraint():
    tablename='public.tablename'
    constname='constraintname'
    try:
        query="ALTER TABLE %s ADD CONSTRAINT %s UNIQUE (amcno,unitid);" 
%(tablename,constname)
        result=db.executesql(query)
    except:
        result="ERROR : constraint already exists"
    finally:
        return locals()
