Red Hat Enterprise Linux 5 aka RHEL 5? You'll be having python 2.4.x installed by default and web2py 2.x.x or newer requires python 2.5 as a minimum. Python 2.6 is available in the Fedora EPEL repository for RHEL 5.
Perhaps you already knew this, but I thought I'd mention it in case you don't... Apart from this, the OS itself shouldn't be a problem.
