Yes, you only need to say the punctuation mark you would like inserted, where you would like to insert it. Inflection or tone has no baring. Here are a few examples; What to say when dictating; What time does the movie start question mark What SIRI will write; What time does the movie start?

As I said, it takes some practice, as it is unnatural to include punctuation within the context of speech.
