Lose the trailing slash.
This always bites people. What you're specifying is a directory _AND_ a
filename prefix, not just a directory.
No more error, but still updates /root/.spamassassin/bayes_stuff

Indeed. It's at least creating the files now. I had created
/etc/mail/spamassassin/bayes_db/bayes
When I removed bayes (as a directory) . . . well . . .ahem.
