That is right you only add the minimum change you need.
In your case the [Service] section wth ExecStartPost=... in it.

Yes it does the merge.

If you want to remove a line from the service file you use
ExecStartPost=
ExecStartPost=...

The first = blank deletes all ExecStartPost in the orginal.
