Oh. I don't think you can hot-deploy a new tiles mapping file; it won't reload it automagically. So you may be asking for a tile definition the system doesn't know about until it's restarted and reloads the tiles.xml file(s).
But really, someone that knows more about this should probably put me in my place before I give you any more suspicious advice!
