This can be done by private vLANs or nested networks, think of it as a
jump box
Initial connection in using Guacamole, then once in on the system, that
system then would have access to the other systems you want.

I believe the answer to your question is No - Guacamole does not
provide a central mechanism by which users can connect to remote
systems using whatever client they like. Guacamole is a remote
access client, not a generic proxy or connection broker for other
clients.
That said, if you choose to provide Guacamole as a way for users
to access remote SSH, RDP, Telnet, etc., systems, it does not mean
that users cannot also use other clients to access those systems.
Guacamole does not do anything to make itself the exclusive client
by which users can access systems - if you wish to achieve this,
you need to use firewalls and proper network architecture to
isolate those systems and funnel access through Guacamole.
