I never heard about a sudden disappearing of the CUPS configuration
files. Can you check whether there is enough disk space for them ("df -h
/etc/cups/")? Do you have /var/log/cups/error_log and /var/log/syslog of
the night where your printers.conf disappeared? Can you post these
files? Is the PPD file in /etc/cups/ppd still present?

If you do not have the logs any more, re-create the printer and as soon
as it disappears again attach the mentioned log files.
