I had the same problem on Ubuntu 9.10 when both VLC and Amarok were open at the same time. Whichever one was opened first would be able to play sound, and the other one would not. I went into both Amarok and KDE4 systemsettings and changed every sound output preference to prefer PulseAudio above all else (which wasn't the default), and then after closing both programs and reopening them, I was able to get both programs to play sound at the same time.
