Start with http://www.qmail.org - it lists a couple reasons you might want/need more than one instance of qmail-send. I ran 2 copies when I was testing out greylisting but didn't want to move it into production. This was years ago though. For the most part, you don't really need more than one instance, since each instance requires another installation of qmail to a different directory (ie. /var/qmail2) and all the processes attached, and each installation has its own queue. It quickly complicates things.
