No, you do not need to be connected to the Internet to use TiddlyWiki under
Node.js. (I frequently run it while travelling internet-less on the train).
Once Node.js is downloaded it doesn't connect to the Internet unless you
run an application that needs to connect to the Internet.

TiddlyDesktop does indeed currently only open standalone TW files.
