Where does SA know that it is supposed to read this file? I guess you need to either tell SA to read the file or to include the configuration statements to your local.cf.
No, you won't need to add lines to local.cf to tell it to read the file.
Spamassassin reads _ALL_ cf files in /etc/mail/spamassassin, not just local.cf.. This is hard-coded behavior, just like reading __prefix__/share/spamassassin/*.cf
