No, do not edit any of the /usr/share/spamassasin/* files if you can avoid it. They will be obliterated when you upgrade.
/etc/mail/spamassassin/local.cf is the proper spot for site-wide applications ~/.spamassassin/user_prefs is the proper spot if you run SpamAssassin as different users and want different settings per user.
