By not having the Python bindings built, you lose the ability to script
GIMP using Python, and hence lose the use of any scripts which are
written in Python. That's pretty much it.

Strangely, Adobe's website lists GIMP as supporting DNG (read-only):

More than its use with GIF, I am glad that the patent on LZW is expiring.
It is a useful dictionary based algorithm that has been used in several
specifications such as PDF, PostScript, TIFF, V.42bis and compress.
