I can confirm this bug in Karmic. The problem does not exist in Jaunty,
so this is a regression.
Could the fix for 42052 or 428115 have caused this regression?

The problem with Ubuntu Tweak is that it totally disables the lock
screen, even when you want it.

gconf-editor only changes key it's not responsive for the use
applications do from those keys later, it's likely that your tweak
software change other keys though
