alternative strategy to plasmid dilution may be inducible expression using the
HEK293S GnTI– TetR cell line (PMID: 12370423) and a compatible expression

In addition to the remarks of Mark and Tim, could you make sure that you are
refining in Refmac with the correct flag for the Rfree set (value = 0)? In
Phenix, this is the opposite: the value is 1 for test reflections and 0 for
work reflections. So going from a PHENIX environment to Refmac, you
SHELXL dropped
R/R-free and improved the GooF and least-squares refinement convergence,
even though the twin fraction was rather low (~ 5%).

can somebody explain to me the exact definitions for detector roll,
pitch and yaw? Can they be detector dependent or rather dependent on the
beamline setup? Is there a relationship with the goniometer position?
