I often, if not always, removed the screws with no ill effects to
derailleur performance. I think that the screws are placed at the dropouts
to allow easy wheel side-to-side alignment, which (assuming your frame is
not wildly out of whack) the ends of the dropouts also do.

Do they have other purposes?

I bought dropout screws for my 2 Riv Road fixies, but I never bothered to
install them, preferring to gauge chain tension and wheel alignment by
thumb and eye -- easy enough!
