My Qmail rc script (/var/qmail/rc) reads like this:

exec env - PATH="/var/qmail/bin:$PATH" qmail-start '|dot-forward .forward
./Maildir/'

Sounds like you are missing the / at the end of "./Maildir", so it's
trying to deliver to a Mailbox.
