You could do it simply with ConfigObj ( 
http://www.voidspace.org.uk/python/configobj.html
):

from configobj import ConfigObj

config = ConfigObj(filename)
section = config['Section Name']
value = section['value']

....

section['value'] = newValue
config.write()
