If you want to do a business application, well, I'd use Seaside and a web based UI. (that's the current conclusion for my own ongoing development)
Otherwise, Spec (which is in a state of flux), or Morphic (which is not covering all you may need in an application). You can also follow the work of Sean De Nigris who is trying to get a Magritte Morphic working.
