Hi Leon, I am sorry I can't help you much with your specific questions, but perhaps you will find my post helpful anyway :)
1) the 2x TC - this is more a question. Wouldn't it be better directly beyond the lens? It's usually the recommended situation. 2) You do 7:1 magnification, right? At such large magnification, I think any small coverage fast lens would cover easily the entire 24x36mm negative! Have you considered an old movie lens, lf shorter focal length? You would get greater DOF too, although with a different angle of view. The C mount lenses for 16mm movie cameras could be useful for this, and can be really cheap as well as plenty fast.
