Hi,

1.  No you do not have to disconnect your Time Machine volume.  I didn't 
disconnect anything when I installed Lion, I had two external Firewire HDs and 
one network volume connected to my Airport Extreme for Time Machine backups and 
there were no issues at all.  Once you choose your install destination, all the 
other connected devices are immaterial.

2.  If you are asking about your Mail preferences and how they point to the 
Mail servers, then the automatic migration during upgrade to Lion will handle 
everything and you shouldn't have to make any changes.  In fact, other than the 
upgrade to Lion, your OS should remember everything that it had in SL.  That 
is, all your preferences, prior server connections, even your Safari history.
